Uncertain significance for Angelman syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_130839.5(UBE3A):c.313T>C (p.Cys105Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 85 of the UBE3A protein (p.Cys85Arg). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with UBE3A-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign". The arginine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
