Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001136139.4(TCF3):c.1787T>C (p.Ile596Thr), citing Invitae Variant Classification Sherloc (09022015): The TCF3 gene has multiple clinically relevant transcripts. This variant occurs in alternate transcript NM_001136139.3, and corresponds to NM_003200.4:c.1823-384T>C in the primary transcript. This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 596 of the TCF3 protein (p.Ile596Thr). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TCF3-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant is not likely to affect RNA splicing. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
