Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.661A>T (p.Met221Leu), citing Ambry Variant Classification Scheme 2023: The p.M221L variant (also known as c.661A>T), located in coding exon 7 of the POLE gene, results from an A to T substitution at nucleotide position 661. The methionine at codon 221 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
