NM_001130987.2(DYSF):c.5087A>G (p.Glu1696Gly) was classified as Uncertain significance for Neuromuscular disease caused by qualitative or quantitative defects of dysferlin by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 1657 of the DYSF protein (p.Glu1657Gly). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with DYSF-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on DYSF protein function. This variant disrupts the p.Glu1657 amino acid residue in DYSF. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 17070050). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing.
