Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.124T>C (p.Ser42Pro), citing Ambry Variant Classification Scheme 2023: The p.S42P variant (also known as c.124T>C), located in coding exon 1 of the CHEK2 gene, results from a T to C substitution at nucleotide position 124. The serine at codon 42 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
