This variant lies in the GJA1 gene (transcript NM_000165.5) at coding-DNA position 679, where G is replaced by C; at the protein level this means replaces glutamic acid at residue 227 with glutamine — a missense variant. Submitter rationale: This sequence change replaces glutamic acid, which is acidic and polar, with glutamine, which is neutral and polar, at codon 227 of the GJA1 protein (p.Glu227Gln). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of oculodentodigital dysplasia (Invitae). In at least one individual the variant was observed to be de novo. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt GJA1 protein function with a positive predictive value of 80%.
