Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003242.6(TGFBR2):c.1567C>G (p.His523Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine, which is basic and polar, with aspartic acid, which is acidic and polar, at codon 523 of the TGFBR2 protein (p.His523Asp). This variant is present in population databases (rs775441401, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with TGFBR2-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt TGFBR2 protein function with a positive predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
