Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000157.4(GBA1):c.115G>C (p.Gly39Arg), citing LabCorp Variant Classification Summary - May 2015: Variant summary: GBA1 c.115G>C (p.Gly39Arg) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. Several computational tools predict a significant impact on normal splicing: Three predict the variant weakens a 5' donor site. One predict the variant creates a 5' donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 5.6e-06 in 1613098 control chromosomes. c.115G>C has been reported in the literature in heterozygous individuals affected with autosomel dominant Parkinsons disease (Crosiers_2016, Zhao_2020, Zhao_2021). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
