This variant lies in the RYR1 gene (transcript NM_000540.3) at coding-DNA position 1931, where G is replaced by T; at the protein level this means replaces arginine at residue 644 with leucine — a missense variant. Submitter rationale: Variant summary: RYR1 c.1931G>T (p.Arg644Leu) results in a non-conservative amino acid change located in the B30.2/SPRY domain (IPR001870) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 250488 control chromosomes. c.1931G>T has been reported in the literature in individuals affected with Myopathy (Klein_2012). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
