This variant lies in the CHST6 gene (transcript NM_021615.5) at coding-DNA position 202, where T is replaced by C; at the protein level this means replaces tyrosine at residue 68 with histidine — a missense variant. Submitter rationale: Variant summary: CHST6 c.202T>C (p.Tyr68His) results in a conservative amino acid change located in the P-loop containing nucleoside triphosphate hydrolases (IPR027417) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 248754 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.202T>C has been reported in the literature in at least an individual affected with macular corneal dystrophy (example: Niel_2003). This report however does not provide unequivocal conclusions about association of the variant with Macular Corneal Dystrophy. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
