This variant lies in the DYNC2H1 gene (transcript NM_001377.3) at coding-DNA position 152, where T is replaced by G; at the protein level this means replaces leucine at residue 51 with arginine — a missense variant. Submitter rationale: Variant summary: DYNC2H1 c.152T>G (p.Leu51Arg) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 249164 control chromosomes. c.152T>G has been reported in the literature in two compound heterozygous fetuses affected with Short-rib thoracic dysplasia (Liu_2021). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
