Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005235.3(ERBB4):c.2017G>A (p.Val673Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 673 of the ERBB4 protein (p.Val673Ile). This variant is present in population databases (rs745465497, gnomAD 0.005%). This variant has not been reported in the literature in individuals affected with ERBB4-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt ERBB4 protein function with a negative predictive value of 80%. Experimental studies have shown that this missense change affects ERBB4 function (PMID: 15534001, 15746097, 24791013). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
