Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001104631.2(PDE4D):c.1029A>C (p.Glu343Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with aspartic acid, which is acidic and polar, at codon 343 of the PDE4D protein (p.Glu343Asp). This variant is present in population databases (rs753598558, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with PDE4D-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt PDE4D protein function with a negative predictive value of 80%. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
