Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000093.5(COL5A1):c.4798A>G (p.Asn1600Asp), citing Ambry Variant Classification Scheme 2023: The p.N1600D variant (also known as c.4798A>G), located in coding exon 62 of the COL5A1 gene, results from an A to G substitution at nucleotide position 4798. The asparagine at codon 1600 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
