Uncertain significance for Developmental and epileptic encephalopathy, 36 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001099922.3(ALG13):c.2645A>G (p.Gln882Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine, which is neutral and polar, with arginine, which is basic and polar, at codon 882 of the ALG13 protein (p.Gln882Arg). This variant is present in population databases (rs763799486, gnomAD 0.02%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with ALG13-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt ALG13 protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
