Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_006129.5(BMP1):c.2075C>T (p.Ser692Phe), citing LabCorp Variant Classification Summary - May 2015: Variant summary: BMP1 c.2075C>T (p.Ser692Phe) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.00022 in 251146 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in BMP1 causing Osteogenesis Imperfecta (0.00022 vs 0.0011), allowing no conclusion about variant significance. c.2075C>T has been reported in the literature in individual(s) affected with bone fragility disorders, without strong evidence for causality (example, Rauch_2019). These report(s) do not provide unequivocal conclusions about association of the variant with Osteogenesis Imperfecta. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
