Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000143.4(FH):c.520C>T (p.Pro174Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with serine, which is neutral and polar, at codon 174 of the FH protein (p.Pro174Ser). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with FH-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on FH protein function. This variant disrupts the p.Pro174 amino acid residue in FH. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 12761039, 16575891, 22069215). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing.
