Uncertain significance for Megalencephaly-polymicrogyria-polydactyly-hydrocephalus syndrome 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005465.7(AKT3):c.382A>G (p.Ile128Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 128 of the AKT3 protein (p.Ile128Val). This variant is present in population databases (no rsID available, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with AKT3-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt AKT3 protein function with a positive predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
