Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004963.4(GUCY2C):c.830+2T>C, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects a donor splice site in intron 6 of the GUCY2C gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in GUCY2C cause disease. This variant is present in population databases (rs780582689, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with GUCY2C-related conditions.
