Pathogenic for Cutis laxa, autosomal recessive, type 1B — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_016938.5(EFEMP2):c.229_230dup (p.Tyr78fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Tyr78Alafs*89) in the EFEMP2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in EFEMP2 are known to be pathogenic (PMID: 17937443). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with EFEMP2-related conditions. For these reasons, this variant has been classified as Pathogenic.
