This variant lies in the NCF2 gene (transcript NM_000433.4) at coding-DNA position 1573 through coding-DNA position 1574, deleting 2 bases; at the protein level this means shifts the reading frame starting at glutamic acid residue 525, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change results in a frameshift in the NCF2 gene (p.Glu525Serfs*23). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 2 amino acid(s) of the NCF2 protein and extend the protein by 20 additional amino acid residues. This variant is present in population databases (rs746336607, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with NCF2-related conditions.
