Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant was absent in 251260 control chromosomes (gnomAD). To our knowledge, there are no reports of c.5521_5528delTTCACCTC in individuals affected with Marfan Syndrome and no experimental evidence demonstrating an impact on protein function. No other clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as likely pathogenic.
