Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015213.4(DENND5A):c.173C>T (p.Thr58Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine, which is neutral and polar, with methionine, which is neutral and non-polar, at codon 58 of the DENND5A protein (p.Thr58Met). This variant is present in population databases (rs140469660, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with DENND5A-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
