Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006361.6(HOXB13):c.800G>A (p.Arg267His), citing Ambry Variant Classification Scheme 2023: The p.R267H variant (also known as c.800G>A), located in coding exon 2 of the HOXB13 gene, results from a G to A substitution at nucleotide position 800. The arginine at codon 267 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
