Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005228.5(EGFR):c.2105C>G (p.Ala702Gly), citing Ambry Variant Classification Scheme 2023: The p.A702G variant (also known as c.2105C>G), located in coding exon 18 of the EGFR gene, results from a C to G substitution at nucleotide position 2105. The alanine at codon 702 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
