Pathogenic for Deficiency of aromatic-L-amino-acid decarboxylase — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001082971.2(DDC):c.853C>T (p.Arg285Trp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 285 of the DDC protein (p.Arg285Trp). This variant is present in population databases (no rsID available, gnomAD 0.06%). This missense change has been observed in individual(s) with aromatic L-amino acid decarboxylase deficiency (PMID: 17533144). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 360433). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt DDC protein function with a positive predictive value of 80%. Experimental studies have shown that this missense change affects DDC function (PMID: 24865461). For these reasons, this variant has been classified as Pathogenic.
