NM_000512.5(GALNS):c.913C>A (p.Pro305Thr) was classified as Uncertain significance for Mucopolysaccharidosis, MPS-IV-A by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015. This variant lies in the GALNS gene (transcript NM_000512.5) at coding-DNA position 913, where C is replaced by A; at the protein level this means replaces proline at residue 305 with threonine — a missense variant. Submitter rationale: The observed missense c.913C>A (p.Pro305Thr) variant in GALNS gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.Pro305Thr variant is absent in gnomAD Exomes. This variant has not been submitted to the ClinVar database. Multiple lines of computational evidence (Polyphen - Probably damaging, SIFT - Tolerated and Mutation Taster - Disease causing) predicts conflicting evidence on protein structure and function for this variant. The reference amino acid at this position on GALNS gene is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. The amino acid Pro at position 305 is changed to a Thr changing protein sequence and it might alter its composition and physico-chemical properties. For these reasons, this variant has been classified as Variant of Uncertain Significance (VUS).
