This variant lies in the CELSR1 gene (transcript NM_001378328.1) at coding-DNA position 6010, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 2004 with asparagine — a missense variant. Submitter rationale: A CELSR1 c.6010G>A (p.Asp2004Asn) variant was identified at a heterozygous allelic fraction of 50.6%, a frequency which may be consistent with germline origin. This variant, to our knowledge, has not been reported in the medical literature. It is observed on 65/1,612,814 alleles in the general population (gnomAD v.4.1.0). Computational predictors are uncertain as to the impact of this variant on CELSR1 function.
