Uncertain significance for Renal coloboma syndrome — the classification assigned by Kasturba Medical College, Manipal, Kasturba Medical College, Manipal, Manipal Academy of Higher Education, Manipal, India to NM_000278.5(PAX2):c.196A>G (p.Ser66Gly), citing ACMG Guidelines, 2015. This variant lies in the PAX2 gene (transcript NM_000278.5) at coding-DNA position 196, where A is replaced by G; at the protein level this means replaces serine at residue 66 with glycine — a missense variant. Submitter rationale: A novel missense variant of uncertain significance, c.196A>G p.(Ser66Gly) in exon 2 of PAX2 (NM_000278.5) was observed in Fetus of proband in heterozygous state. On segregation, this variant was observed in heterozygous state in proband and DNA of the first aborted fetus (Lab ID: WES24324) as well. Wild type allele was observed in her husband, (Lab ID: WES 24256). This variant is absent in the gnomAD (v4.1.0) population database and in our in-house data of 3455 exomes.
