Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001127898.4(CLCN5):c.673G>A (p.Val225Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 155 of the CLCN5 protein (p.Val155Ile). This variant is present in population databases (rs200390605, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with CLCN5-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on CLCN5 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
