NM_001277115.2(DNAH11):c.5318A>G (p.His1773Arg) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H1773R variant (also known as c.5318A>G), located in coding exon 30 of the DNAH11 gene, results from an A to G substitution at nucleotide position 5318. The histidine at codon 1773 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious bySIFT in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
