Benign — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000089.4(COL1A2):c.2082C>A (p.Gly694=), citing LabCorp Variant Classification Summary - May 2015: Variant summary: COL1A2 c.2082C>A (p.Gly694Gly) alters a non-conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 0.00013 in 250778 control chromosomes. The observed variant frequency exceeds the estimated maximal expected allele frequency for disease-causing variants in COL1A2. To our knowledge, no occurrence of c.2082C>A in individuals affected with COL1A2-related conditions and no experimental evidence demonstrating its impact on protein function have been reported.
