This variant lies in the FAT4 gene (transcript NM_001291303.3) at coding-DNA position 6580, where G is replaced by A; at the protein level this means replaces valine at residue 2194 with isoleucine — a missense variant. Submitter rationale: A FAT4 c.6580G>A (p.Val2194Ile) variant was identified at a near heterozygous allelic fraction of 48.5%, a frequency which may be consistent with germline origin. This variant, to our knowledge, has not been reported in the medical literature. It is only observed on 3/1,613,962 alleles in the general population (gnomAD v.4.1.0), indicating it is not a common variant. Computational predictors suggest that the variant does not impact FAT4 function.
