NM_004415.4(DSP):c.3601G>A (p.Glu1201Lys) was classified as Uncertain Significance for Arrhythmogenic cardiomyopathy with wooly hair and keratoderma by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 3601, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 1201 with lysine — a missense variant. Submitter rationale: This missense variant replaces glutamic acid with lysine at codon 1201 of the DSP protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with arrhythmogenic right ventricular cardiomyopathy (PMID: 24125834), as well as in a healthy individual (PMID: 29618732). This variant has been identified in 2/250966 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
