NM_000433.4(NCF2):c.855G>C (p.Gln285His) was classified as Pathogenic for Granulomatous disease, chronic, autosomal recessive, cytochrome b-positive, type 2 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine, which is neutral and polar, with histidine, which is basic and polar, at codon 285 of the NCF2 protein (p.Gln285His). RNA analysis indicates that this missense change induces altered splicing and may result in an absent or altered protein product. This variant is present in population databases (no rsID available, gnomAD 0.002%). This missense change has been observed in individual(s) with chronic granulomatous disease (PMID: 32666379). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 3574675). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Studies have shown that this missense change results in skipping of exon 8, and produces a non-functional protein and/or introduces a premature termination codon (PMID: 32666379). For these reasons, this variant has been classified as Pathogenic.
