Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_016169.4(SUFU):c.71dup (p.Ala25fs), citing Ambry Variant Classification Scheme 2023: The c.71dupC (p.A25Gfs*23) alteration, located in exon 1 (coding exon 1) of the SUFU gene, consists of a duplication of C at position 71, causing a translational frameshift with a predicted alternate stop codon after 23 amino acids. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. The SUFU c.71dupC alteration was flagged as a low confidence call in the Genome Aggregation Database (gnomAD). This variant has been reported in four patients diagnosed with medulloblastoma before 3 years of age from a large family (Brugières, 2010). Based on the available evidence, this alteration is classified as pathogenic.
