Pathogenic — the classification assigned by Ambry Genetics to NM_015902.6(UBR5):c.3060dup (p.Ile1021fs), citing Ambry Variant Classification Scheme 2023: The c.3060dupC (p.I1021Hfs*5) alteration, located in coding exon 23 of the UBR5 gene, consists of a duplication of C at position 3060, causing a translational frameshift with a predicted alternate stop codon after 5 amino acids. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on the available evidence, this alteration is classified as pathogenic.
