NM_000518.5(HBB):c.15T>A (p.Thr5=) was classified as Uncertain significance by Quest Diagnostics Nichols Institute San Juan Capistrano, citing Quest Diagnostics criteria: The HBB c.15T>A (p.Thr5=) synonymous variant (also known as CD 4 (ACT>ACA)) has been reported in the published literature in individuals affected with beta thalassemia that also carry other HBB variants including HBB c.16C>T (p.Pro6Ser) and HBB c.19G>T (p.Glu7*) (PMID: 9371533 (1997), 11074564 (2000)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Analysis of this variant using software algorithms for the prediction of the effect of nucleotide changes on splicing yielded predictions that this variant does not affect HBB mRNA splicing. Based on the available information, we are unable to determine the clinical significance of this variant.
