NM_006502.3(POLH):c.-5+3A>G was classified as Uncertain significance for Xeroderma pigmentosum by Sema4, Sema4, citing Sema4 Curation Guidelines. This variant lies in the POLH gene (transcript NM_006502.3) at 3 bases into the intron immediately after 5 bases upstream of the translation start (5' untranslated region), where A is replaced by G. Submitter rationale: The POLH c.-5+3A>G variant has not been reported in the literature to our knowledge. It was observed in 21/1560 chromosomes of the East Asian cohort in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID: 356897). In silico tools developed to predict the effect of sequence changes on RNA splicing do not suggest a negative effect on normal splicing, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
