NM_001378454.1(ALMS1):c.1865A>G (p.Tyr622Cys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y623C variant (also known as c.1868A>G), located in coding exon 8 of the ALMS1 gene, results from an A to G substitution at nucleotide position 1868. The tyrosine at codon 623 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is well conserved on limited sequence alignment. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
