This variant lies in the ABCC8 gene (transcript NM_000352.6) at coding-DNA position 4368, where C is replaced by G; at the protein level this means replaces isoleucine at residue 1456 with methionine — a missense variant. Submitter rationale: Variant summary: ABCC8 c.4368C>G (p.Ile1456Met) results in a conservative amino acid change located in the ABC transporter-like, ATP-binding domain (IPR003439) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 224262 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.4368C>G in individuals affected with Neonatal Diabetes Mellitus and no experimental evidence demonstrating its impact on protein function have been reported. Two clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 and classified the variant as likely pathogenic, and as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
