Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant was absent in 251576 control chromosomes. c.3545G>A has been observed in individuals affected with Neonatal Diabetes Mellitus (Babenko_2006, Vaxillaire_2007, Flanagan_2007, Alves_2012). These data indicate that the variant is likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function (Ortiz_2012), however, does not allow convincing conclusions about the variant effect.
