NM_002382.5(MAX):c.30G>C (p.Glu10Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E10D variant (also known as c.30G>C), located in coding exon 1 of the MAX gene, results from a G to C substitution at nucleotide position 30. The glutamic acid at codon 10 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species; however, aspartic acid is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
