Pathogenic for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_013275.6(ANKRD11):c.2982dup (p.Lys995fs), citing Ambry Variant Classification Scheme 2023: The c.2982dupG (p.K995Efs*23) alteration, located in exon 9 (coding exon 7) of the ANKRD11 gene, consists of a duplication of G at position 2982, causing a translational frameshift with a predicted alternate stop codon after 23 amino acids. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on the available evidence, this alteration is classified as pathogenic.
