NM_170707.4(LMNA):c.950A>G (p.Glu317Gly) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E317G variant (also known as c.950A>G), located in coding exon 6 of the LMNA gene, results from an A to G substitution at nucleotide position 950. The glutamic acid at codon 317 is replaced by glycine, an amino acid with similar properties. Another variant at the same codon, p.E317K (c.949G>A), has been reported in association with dilated cardiomyopathy and dysrhythmia (Arbustini E et al. J. Am. Coll. Cardiol. 2002;39:981-90; Villa F et al. Immun Ageing. 2014;11:19). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
