This variant lies in the SLC39A4 gene (transcript NM_130849.4) at coding-DNA position 970 through coding-DNA position 973, deleting 4 bases; at the protein level this means shifts the reading frame starting at serine residue 324, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: A known frameshift deletion, c.970_973del in exon 5 of SLC39A4 was observed in homozygous state in the proband (Küry et al., 2002; VCV000003535.1). This variant is absent in heterozygous and/or homozygous state in gnomAD (v4.1.0) and in our in-house database of 4019 exomes. This frameshift deletion is predicted to introduce a premature termination codon in the transcript which may either lead to nonsense-mediated mRNA decay or the formation of a truncated protein product. The clinical features observed in the proband are in concordance with acrodermatitis eneteropathica. Thus, the above-mentioned variant in homozygous state in the proband confirms the diagnosis of acrodermatitis enteropathica in her.
