NM_001365951.3(KIF1B):c.4025A>G (p.Lys1342Arg) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K1296R variant (also known as c.3887A>G), located in coding exon 35 of the KIF1B gene, results from an A to G substitution at nucleotide position 3887. The lysine at codon 1296 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
