Uncertain significance — the classification assigned by Ambry Genetics to NM_052947.4(ALPK2):c.1079A>G (p.Asp360Gly), citing Ambry Variant Classification Scheme 2023: The p.D360G variant (also known as c.1079A>G), located in coding exon 3 of the ALPK2 gene, results from an A to G substitution at nucleotide position 1079. The aspartic acid at codon 360 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
