Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000214.3(JAG1):c.127C>G (p.Gln43Glu), citing Ambry Variant Classification Scheme 2023: The p.Q43E variant (also known as c.127C>G), located in coding exon 2 of the JAG1 gene, results from a C to G substitution at nucleotide position 127. The glutamine at codon 43 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
