Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.9620A>G (p.Glu3207Gly), citing Ambry Variant Classification Scheme 2023: The p.E3208G variant (also known as c.9623A>G), located in coding exon 11 of the ALMS1 gene, results from an A to G substitution at nucleotide position 9623. The glutamic acid at codon 3208 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
